چکیده
UNLABELLED The protein-protein interactions between viral and host proteins play an essential role in plant virus infection and host defense. The potyviral nuclear inclusion protein a protease (NIa-Pro) is involved in various steps of viral infection. In this study, the host proteins interacting with papaya ringspot virus (PRSV) NIa-Pro were screened in a Carica papaya L. plant cDNA library using a Sos recruitment two-hybrid system (SRS). We confirmed that the full-length EIF3G, FBPA1, FK506BP, GTPBP, MSRB1, and MTL from papaya can interact specifically with PRSV NIa-Pro in yeast, respectively. These proteins fufill important functions in plant protein translation, biotic and abiotic stress, energy metabolism and signal transduction. In this paper, we discuss possible functions of interactions between these host proteins and NIa-Pro in PRSV infection and their role in host defense. KEYWORDS Sos recruitment two-hybrid system; papaya ringspot virus; NIa-Pro; protein-protein interaction.

Cloning of RPL10A from Kenaf and studies of its interactions with Hibiscus Chlorotic Ringspot Virus coat protein using yeast-two-hybrid and pull down assays
The focus of viral research in recent years have been pointed towards a better understanding of the mechanisms of plant viral infection and methods to prevent plant viral infections. Hibiscus chlorotic ringspot virus (HCRSV) coat protein (CP) is required for encapsidation and viral systemic movement. Previous studies using affinity chromatography has identified a 60S ribosomal protein L10A (RPL...
متن کاملGeneration of a Papaya Hybrid Variety with Broad-Spectrum Transgenic Resistance to Papaya ringspot virus and Papaya leaf-distortion mosaic virus
The aphid-borne potyvirus, Papaya ringspot virus (PRSV), is the major limiting factor for papaya production worldwide. Transgenic resistance conferred by the PRSV-coat protein (CP) gene and based on the mechanism of post-transcriptional gene silencing (PTGS), has become the most effective method to protecting papaya from infection by the noxious virus. The PRSV-CP transgenic lines of cultivars ...
متن کاملDiscovering Domains Mediating Protein Interactions
Background: Protein-protein interactions do not provide any direct information regarding the domains within the proteins that mediate the interactions. The majority of proteins are multi domain proteins and the interaction between them is often defined by the pairs of their domains. Most of the former studies focus only on interacting domain pairs. However they do not consider the in...
متن کاملInhibition of the Host Proteasome Facilitates Papaya Ringspot Virus Accumulation and Proteosomal Catalytic Activity Is Modulated by Viral Factor HcPro
The ubiquitin/26S proteasome system plays an essential role not only in maintaining protein turnover, but also in regulating many other plant responses, including plant-pathogen interactions. Previous studies highlighted different roles of the 20S proteasome in plant defense during virus infection, either indirectly through viral suppressor-mediated degradation of Argonaute proteins, affecting ...
متن کاملPapaya ringspot virus-P: characteristics, pathogenicity, sequence variability and control.
TAXONOMY Papaya ringspot virus (PRSV) is an aphid-transmitted plant virus belonging to the genus Potyvirus, family Potyviridae, with a positive sense RNA genome. PRSV isolates belong to either one of two major strains, P or W. The P strains infect both papaya and cucurbits whereas the W strains infect only cucurbits. GEOGRAPHICAL DISTRIBUTION PRSV-P is found in all major papaya-growing areas....
